Dilution = initial volume / final volume

Final volume = initial volume + volume of diluent

Dilution = 1/4

Final volume = 800 uL

1/4 = initial volume / 800

Initial volume = 800/4 = 200 uL

Volume of diluent = 800 - 200 = 600 uL

So, answers are 200 and 600 respectively.
