Often a protein modification acts as a signal for cell processes to occur. Such modification act as stimulus for the downstream pathway and avoid false reactions just in presence of protein and not the modifier.

Eg:

-receptor tyrosine kinases undergo autophosphorylation on binding ligand. phosphorylated receptor then signals downstream pathway

-proteins are ubiquitinated, so they are tagged for proteolysis

-often methylated proteins have had increased/attenuated function, have implications in certain diseases
