Basic dye means, it is either able to accept H+ ion or release OH- ion into an aqueous solution, so, that it becomes positively charged. We say that it is basic in nature because it interacts with bacterial cell wall which is negatively charged.

(Acidic dyes are negatively charged and interact with positively charged structures in a cell).

To confirm that safranine is a basic dye, we can perform chemical test. Basic chemicals turn red litmus blue. Also, by using pH meter, we can check if the pH of safranine is more than 7 or not. Both of these tests confirm that safranine is basic in nature.
