Answer : Restriction enzymes are used in

  1. Genetic finger printing
  2. Genetic engineering
  3. Insulin production using E coli

Explanation : DNA replication do not require any restriction enzymes as it is a in Vivo process which requires DNA polymerase III in prokaryotes and DNA polymerase delta in eukaryotes which are the prime DNA synthesizing enzymes. Further, Polymerase chain reaction also do not require these enzymes as PCR involves only the amplification of given sample of DNA which requires heat stable DNA polymerase such as Taq polymerase (obtained from Thermus aquaticus), all four dNTPs and Two primers (Forward and Reverse primers).  

Other techniques that involves restriction enzymes includes, Restriction fragment length polymorphism (RFLP technique), DNA mapping, cDNA construction etc.,

  • Eco R1 obtained from E coli
  • Hind III from Haemophilus influenzae
  • Bam H1 from Bacillus amyloliquifaciens etc.,
