More closely related the organisms are , there is more similarity between their DNA sequences. In a hybridisation experiment , we use DNA probes that are complimentary to the test sample. Suppose we have the DNA probe from a species, and we have to check if out sample is related. We conduct hybridisation experiment with the probe that we have.

This hybridisation prove binds to the complimentary strand of the DNA sample. If the DNA sample sequence and probe sequence are similar , they bind to each other properly giving a nice double strand. If yhe sequence are not similar , there will be loops and bulges in places where the variation is there.
