The linear concentration range for the Bradford assay is 20-2000 µg/mL of protein. Protein concentrations below or above these ranges cannot be measured accurately. If it is suspected that a sample contains greater than 50 mg/mL of protein, how can the protein concentration still be measured using the Bradford assay? Explain.

Answer #1

Ans- if sample protein concentration is greater then linear range than you would need to dilute the sample.

In this question, the maximum linear range is 2 mg/ml, and your sample contains protein concentration 50mg/ml,

then you dilute your sample 100 times.

It becomes 0.5mg now; you can easily measure the protein concentration of your sample. 100 time dilution would be 99 part of your choice of buffer and 1 part of your sample.

To calculate the concentration of the undiluted, unknown sample, simply multiply by the dilution factor. 100*0.5=50 mg
