The conversion of glyceraldehyde 3-phosphate to 1,3-bisphosphoglycerate is catalyzed by the enzyme glyceraldehyde 3-phosphate dehydrogenase. Formation of the enzyme-substrate complex involves nucleophilic attack by an active site cysteine residue on glyceraldehyde 3-phosphate to form a tetrahedral intermediate. However, cysteine-dependent nucleophilic attack only occurs when NAD+ is bound, which depresses the pKR for cysteine from 8 to 5.5. Please propose an alternative active site arrangement that may lead to the formation of a tetrahedral covalent intermediate without the requirement for NAD+ (at pH = 7.4).
