Question

A patient was presented with suspected salicylic acid (SA) overdose. The initial plasma concentration of SA was 2000 mg/L and 1 hour later it is 1000 mg/L.

a) What is the half-life of SA in this patient? (2)

b) Calculate the elimination constant of SA in this patient. (4)

c) Calculate how many hours will be needed for the SA to reach a level of 62.5 mg/L if the kel = 0.693 in this patient. (answer 5 hours) (4)

You may find the following equations useful: LogCt = Log Co – (kel X t)/2.303; Half-life (t1/2) = 0.693/kel; Vd = dose (mg) / plasma concentration (mg/L); % Bioavailability = (AUCoral/AUCiv)x100 Vd = Dose i.v. / (AUC x kel)

    1. A new drug was given in a single intravenous dose of 200 mg to an 80- kg adult male patient. The apparent VD is 0.8 L/kg, and the drug is eliminated by 1st order kinetics. Compute the initial drug plasma concentration. 2. A drug has an elimination half-life of 4 hours and follows first-order kinetics. An adult male (73 years old, 65 kg) with diabetes mellitus is placed on hemodialysis. His residual creatinine clearance is <5mL/min. The patient is given tobramycin, an aminoglycoside antibiotic, at a dose, at a dose of 1 mg/kg by IV bolus injection. Tobramycin is 90% excreted unchanged in the urine, is less than 10% to plasma protein, and has elimination half-life 2.2 hours in patient with normal renal function. In this patient, tobramycin has an elimination t1/2 of 50hours during...

If the average observed AUC after administration of 300 mg IV theophylline in nonsmokers is 100 mg.hr/L, what is the expected AUC in smokers after administration of the same 300 mg dose? Theophylline undergoes linear pharmacokinetics 2. Please show work. A 86.5 kg patient has just received a single 500 mg oral dose of an antibiotic. This antibiotic has a 90% oral bioavailability, 8.6 hour elimination half-life, and a 1.1 hour absorption half-life. The drug literature states that the population volume of distribution is 1.1 L/kg for this medication. A blood sample was collected at the maximum drug concentration and reported to be 5.916 mg/L. What is the patient-specific volume of distribution of this antibiotic in this...
