In shotgun sequencing overlapping DNA segments are sequenced and then assembled to form the whole sequence. It works best for genomes with less repeat sequences.

In this method the DNA is sheared into smaller fragments and used to prepare a shotgun library. A shotgun library has multiple plasmid vector into which each DNA fragment is cloned. Universal primers are used to sequence each DNA fragment in the shotgun library. The sequences usually there are overlaps between the fragments which is then used to assemble them in order to get the sequence of the whole genome.

Each fragment has been colour coded for easy identification. Only the novel sequences from each fragment is added to the final sequence.
