Exome sequencing only identifies protein coding genes (exons); it does not identify other non-protein coding genes (introns). A majority of known disease causing mutations are found in or adjacent to the exons. While preparing the genomic DNA (deoxyribonucleic acid) for exome sequencing, only protein coding genes are isolated, and non-coding genes are skipped. So, disease causing mutations cannot be identify by exome sequencing.
