Exome sequencing is a molecular laboratory technique used to sequence all the protein coding genes (exons) in a genome. In order to sequence only exons in the genome, genomic DNA (deoxyribonucleic acid) is first cut into small pieces (fragmentation), and then denatured. Then the denatured fragments are hybridized with exon specific biotin-labelled probes. These probes specifically bind to the exons, so duplexes containing annealed probes are isolated and prepared for whole exome sequencing.
